白及微卫星引物筛选及群体遗传多样性研究

  白及(Bletilla striata Rchb.)是我国二级保护植物,也是重要的药用植物,野生资源枯竭,亟需保护.本研究利用改良CTAB法提取分离白及的基因组DNA,利用筛选的白及多态性微卫星引物对白及4个野生居群80个样品进行遗传多样性分析.筛选出的20对白及微卫星引物能够成功扩增样品的DNA;这些引物扩增产物的等位基因数目为2~6个,观察杂合度范围为0.3500~0.7000,期望杂合度范围为0.4685~0.7653,多态性信息含量范围为0.3572~0.7229;使用4个白及近缘种(华白及、小白及、黄花白及、绶草)进行引物通用性检测,20对引物能够在4个物种中成功扩增.因此,筛选出的多态性微卫星引物可以用于白及的遗传多样性和群体遗传结构研究.
