Cows milk is one of the most common allergenic foods,especially in infant and children,and β-lactoglobulin(BLG)is the main allergen in cows milk.The traditional method to detect BLG is enzyme-linked immunosorbent assay(ELISA).However,in the conventional ELISA experiment,antibodies binding to rabbit or mouse IgG epitopes of BLG,not human epitopes of BLG.Here,we used a new kind of antibody which recognized human IgE linear epitopes of BLG,and a sandwich ELISA was developed based on this antibody and rabbit anti-BLG polyclonal antibody.The linear range for BLG detection was 31.25 ng/mL to 8 × 103 ng/mL,the limit of detection and quantitation were 1.96 ng/mL and 3.91 ng/mL,respectively.The relative standard deviation of repeatability(RSDr)and reproducibility(RSDR)were 10.66%and 11.72%,respectively.In addition,this method had a good specificity as well,except a slight cross-reactivity with caseins.Five kinds of hydrolysed formulas has been evaluated,and BLG allergic residues could be detected.Compared to the traditional method,this assay not only can detection and quantitation of BLG in milk samples,but can detection of BLG allergic residues in hydrolysed formulas and other processed milk products.
