乳鼠直结肠扩张后在体骨髓干细胞自发分化表达神经细胞相关基因初探

  目的 骨髓干细胞可以诱导分化成神经细胞,TRPV1是瞬时受体电位通道家族中最重要的成员之一,在胃肠病及与其相关的内脏高敏感中有重要的作用,新生动物进行直结肠扩张会引起内脏痛敏,本实验目的是探讨新生动物引起内脏痛敏过程中是否有骨髓干细胞自发分化为神经细胞和TRPV1阳性细胞参与.方法 选用新生SD大鼠,随机分为实验组和对照组,在生后第8~14天实验组进行直结肠扩张,制作内脏痛敏模型,第15天实验组与对照组取全骨髓细胞作RT-PCR.按照RNA提取试剂盒说明书提取总RNA.以mRNA作为模板进行逆转录,PCR扩增.以GAPDH为内参照,检测TRPV1 (PCR产物长度262 bp)、CD34 (199 bp)、c-kit (327 bp)、nesting (381 bp)、β-tubulin (246 bp)、GFAP (437 bp) mRNA的表达.PCR产物经1.5%琼脂糖凝胶电泳后进行光密度扫描,将目的扩增条带的光密度值与GAPDH的光密度值之比作为衡量参数,分析各组mRNA的表达.结果 验组神经干细胞或前体细胞标志物nesting、β-tubulin高表达,干细胞标志物CD34、c-kit没有表达,与内脏痛敏有关的TRPV1弱表达,对照组CD34、c-kit、nesting、β-tubulin均有表达,TRPV1几乎无表达,实验和对照组中均未见星型胶质细胞标志物(GFAP)蛋白的表达.结果提示部分CD34、c-kit阳性细胞在体内脏痛敏条件下可能自发分化为神经干细胞或前体细胞以及TRPV1阳性细胞. 结论 骨髓干细胞在新生大鼠体内经直结肠扩张后能自发分化为与内脏痛敏有关的神经细胞和TRPV1阳性细胞,骨髓干细胞可能参与内脏痛敏的发展进程.
