  [Objective]Tetrodotoxin(TTX) is a highly toxic low-molecular and non-protein marine biotoxin,and it is also one of the most toxic neurotoxin in nature.Solid-phase microextraction(SPME) is a new extraction technique which is based on the solid-phase extraction(SPE).It is a system which includes sampling,extraction,concentration,with the advantage of simple operation,rapid,low cost,safety,good choice,high sensitive and solventlessness.To verify the possibility that tetrodotoxin could be extracted from its standard using solid-phase microextraction and determine its extraction efficiency,the experiment was conducted.[Methods]First,the tetrodotoxin standard was diluted by mobile phase which was made of water,acetic acid and tetrahydrofuran(v∶v∶v=1000∶1.5∶5),forming solutions with three different concentrations(40mg/L、 20mg/L and 10mg/L).Then solid-phase microextraction and elution were conducted in bottles under ambient temperature,which lasted 30min and 20min respectively.The SPME fiber used was 85μm polyacrylate(PA).Finally,the absorbance values of TTX at 196.1 nm were obtained from UV spectrophotometer.[Results]Calibration curve was linear over the TTX concentration range of 15~40mg/L(y=0.005x+ 1.589,r=0.9654).[Conclusion]It was concluded that TTX could be extracted from its standard using solid-phase microextraction,but the extraction efficiency was not satisfactory enough due to the short extraction and elution time.
