ATRX knockdown changed ALT and increased stemness in glioma

  More than 30%malignant glioma will maintain the telomere length by the mechanism of alternative lengthening of telomere(ALT),but the specific mechanism of ALT is unclear.ATRX is a chromatin remodeling complex protein.Our recent studies have shown that ATRX mutation is related with ALT in brain tumors.ATRX gene was knockdown in different glioma cell lines with RNAi to study its tumorgenesis effects.Our experiments showed that the loss of ATRX changed the phenotype of ALT in JHH-GBM14 cell line by FISH staining; and that the loss of ATRX also significantly increased the expression of stemness markers in JHH-GBM14 and JHH-040821 cell lines.Our study proved that ATRX gene may play an important role in glioma ALT phenotype.It is suggested that ATRX may drive the tumor biological characteristics formation including ALT and stemness mechanism in brain tumor.
