新型扩张床中介质及蛋白吸附的轴向分布研究

  扩张床吸附(Expanded bed adsorption)是一种新型生物分离方法,把下游过程中的固液分离、浓缩和初步纯化集成于一个单元操作,减少操作步骤,提高产品收率,降低分离费用,在蛋白质、抗体、酶及基因工程药物的分离纯化中具有广阔的应用前景.扩张床吸附最重要的特征是床层内稳定的轴向分级分布,本文针对新型喷嘴型扩张床,采用在线取样方法,比较不同介质和蛋白吸附的轴向分布特征,为设计新型扩张床及介质提供理论依据.首先,考察了介质在扩张床内的轴向分布,包括介质的粒径、密度及床层空隙率随床层高度的变化.随床层高度增加,介质粒径减小,粒径分布逐渐变窄;介质密度减小,床层空隙率逐渐增大.入口处空隙率大约0.65~0.75,顶部空隙率约为0.85.其次,利用停留时间分布考察床层内的液流分布,发现床层入口附近流体混合与轴向分散较强烈,入口效应明显.随着床层高度的增加,流动接近平推流,流体混合和轴向分散减弱.最后,考察了蛋白吸附沿床层轴向的变化,在不同床层高度取样测定蛋白穿透曲线,发现蛋白吸附同样存在轴向分布,底部区域对蛋白的吸附贡献最大.结果表明,随着床层高度变化,扩张床各项参数均发生规律性变化,包括介质粒径和密度、床层空隙率、流体混合和蛋白质吸附等.比较分析不同液流分布器和介质的轴向分布特征,可以为新型扩张床及介质设计提供一定的理论指导.
