  Protein samples present a difficult problem in analytical chromatography.Very small differences in chemical structure can have a large impact on biological activity, and a single protein can harbor one or more modification.Further, a sample is essentially a family of modified structures, usually at low concentrations, that are derived from a single native structure.The analytical method must, therefore, detect and quantitate small structural changes on a very large molecule.This problem requires a different approach to chromatographic analysis than used for small molecule pharmaceuticals.The most useful approach to these analyses is the application of chromatographic tools that are each very specific for one chemical property.The sample may be analyzed using an appropriate combination of the techniques of reversed phase, ion exchange, and size exclusion.We will describe an automated system approach that facilitates the free choice of analytical separation method for a particular sample and also permits the analysis of a single sample with all the techniques in series.A column manager directs the sample to the required column while solvent selection valves match the mobile phase constituents.
