目的 :观察中药益髓生血灵对小鼠骨髓造血祖细胞的刺激作用 ,以探讨其治疗 β地中海贫血的疗效机理。方法 :以益髓生血灵 1 .9、3 .8和 7.6g· kg· d- 13个剂量分别给小鼠灌胃 ,并分别以生理盐水、环磷酰胺和生血丸对照组 ,1 0天后应用半固体琼脂克隆形成法及微量甲基纤维素法 ,研究小鼠骨髓粒单系祖细胞( CFU-GM)和早期红系祖细胞 ( BFU-E)和晚期红系祖细胞 ( CFU-E)的生长情况。结果 :益髓生血灵灌胃的 3个剂量组的小鼠骨髓 CFU-GM、BFU-E和 CFU-E集落数都明显增多 ,与生理盐水对照组比较 CFU-GM和 CFU-E集落数的增多有统计学意义 ( P<0 .0 1 ) ,1 .9g· kg· d- 1和 3 .8g· kg· d- 1剂量组的 BFU-E集落数增多也有统计学意义 ( P<0 .0 5和 P<0 .0 1 )。结论 :益髓生血灵对小鼠骨髓 GM-CFU、 BFU-E和 CFU-E均有促进增殖分化作用。
Objective: To observe the stimulatory effect of Chinese herbal medicine Yixu Shengxueling on bone marrow hematopoietic progenitor cells in mice to explore the therapeutic mechanism of β-thalassemia. METHODS: Mice were given intragastric administration of 1.9, 3, 8 and 7.6 g·kg·d - 13 doses of Yishenshengxueling respectively, and they were administered saline, cyclophosphamide and Shengxue Pills respectively, after 10 days. Semiquantitative agar cloning method and trace methylcellulose method were used to study mouse bone marrow mononuclear progenitor cells (CFU-GM) and early erythroid progenitor cells (BFU-E) and late erythroid progenitor cells (CFU-E). ) The growth situation. RESULTS: The number of CFU-GM, BFU-E and CFU-E colonies in the three doses of mice fed with Yigu Shengxueling was significantly increased, and the number of CFU-GM and CFU-E colonies was compared with that of saline control group. The number of BFU-E colonies increased statistically (P<0.01), 1.9g.kg.d-1 and 3.8g.kg.d-1 doses also had statistical significance (P<0. .0 5 and P<0.0 1 ). Conclusion : Yigu Shengxueling can promote the proliferation and differentiation of mouse bone marrow GM-CFU, BFU-E and CFU-E.
