目的:探讨水蛭、何首乌、丹参、三七、赤芍、水蜈蚣等6味中药防治家兔髂动脉球囊扩张术后再狭窄模型的作用机理。方法:采用手术加高脂饲料建立家兔髂动脉球囊扩张术后再狭窄模型,观察上述6味药供试品(约为临床人常用量的4倍等效剂量)于造模同时连续灌胃给药6周后对家兔血清同型半胱氨酸(Hcy)、髂动脉血管组织中白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)及超氧化物歧化酶(SOD)、丙二醛(MDA)等的影响。结果:与模型组比较,水蛭、丹参、水蜈蚣组家兔血清Hcy显著降低(P<0.05或P<0.01),丹参、何首乌组家兔髂动脉血管组织中TNF-α显著降低(P<0.05),水蛭、丹参、水蜈蚣、何首乌组血管组织中SOD活力显著升高(P<0.05或P<0.01),水蛭、丹参、水蜈蚣组MDA显著降低(P<0.05或P<0.01)。结论:降低家兔血清Hcy、血管组织中TNF-α及抗氧化(升高血管组织中SOD、降低MDA)可能是水蛭等中药防治家兔髂动脉球囊扩张术后再狭窄部分作用机制。
Objective: To investigate the mechanism of six kinds of traditional Chinese medicines such as Radix Astragali, Polygonum multiflorum, Radix Salviae Miltiorrhizae, Panax Notoginseng, Radix Paeoniae Rubra and Radix Astragali in preventing and treating restenosis after balloon angioplasty in rabbits. METHODS: The model of restenosis after balloon angioplasty in rabbits was established by surgery and high-fat diet. The above-mentioned 6 herbal medicines (about 4 times the equivalent dose used by the clinician) were observed at the same time for continuous irrigation. After 6 weeks of gastric administration, serum homocysteine (Hcy), iliac artery vascular tissue interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) and superoxide dismutase ( Effects of SOD), malondialdehyde (MDA), etc. RESULTS: Compared with the model group, the serum Hcy levels in the rabbits of the water blister, salvia miltiorrhiza and leechi group were significantly lower (P<0.05 or P<0.01), and the levels of TNF-alpha in the iliac artery of the rabbits of the Danshen and Heshouwu groups were significantly lower (P<0.05). The activity of SOD in the vascular tissues of Shuizhan, Salvia miltiorrhiza, Shuizhi, and Polygonum multiflorum was significantly increased (P<0.05 or P<0.01), and the MDA in the leech, salvia miltiorrhiza, and leeches was significantly decreased (P<0.05 or P<0.01). Conclusion: Decreasing serum Hcy, TNF-α in vascular tissue and anti-oxidation (increasing SOD in vascular tissue and decreasing MDA) in rabbits may be the mechanisms of action of Chinese herbal medicines such as leech, etc. on restenosis after balloon angioplasty in rabbits.
