目的:研究D3S1358基因座两种引物在同一模板不同降解时间扩增效果,对其在法医学上的应用进行可行性探讨。方法:从商品化STR试剂盒中筛选出D3S1358,提取32例无关男性血液样本进行降解、扩增、琼脂糖电泳等。结果:通过琼脂糖电泳结果分析,D3S1358降解后由miniSTR扩增效果明显优于常规STR扩增效果。结论:miniSTR在处理腐败降解检材的分型检出率较常规STR有一定的优势。
OBJECTIVE: To study the amplification effect of two primers of D3S1358 locus at different degradation time in the same template, and to explore its feasibility in forensic science. Methods: D3S1358 was screened from commercial STR kit and 32 unrelated male blood samples were extracted for degradation, amplification and agarose electrophoresis. Results: The result of agarose gel electrophoresis showed that the amplification result of miniSTR by D3S1358 was better than that of conventional STR. Conclusion: The detection rate of miniSTR in treating spoilage samples is higher than that of conventional STR.
