目的:采用高效液相色谱质谱联用(LC-MS/MS)方法测定卷柏药材中的熊果苷的含量。方法:色谱柱为Diamonsil C18(5μm,150 mm×4.6 mm,i.d.)色谱柱,流动相为为乙腈-0.4%甲酸(80∶20,v/v),流速为0.6 m L·min-1,柱温为室温,进样量为10μL;采用电喷雾离子源进行正离子模式检测,选择性反应检测模式(SRM)用于定量测定。结果:熊果苷在1.0~150μg·m L-1范围内与峰面积线性关系良好,低、中、高3个水平的加样回收率分别为98.8%、98.8%和99.7%,RSD均小于1.1%。结论:该方法简便,准确,重现性好,可用于卷柏中熊果苷的含量测定和质量控制。
Objective: To determine the content of arbutin in Selaginella tamarisse by high performance liquid chromatography-mass spectrometry (LC-MS / MS). METHODS: The chromatographic column was Diamonsil C18 (5 μm, 150 mm × 4.6 mm, id). The mobile phase was acetonitrile-0.4% formic acid (80:20, v / v) and the flow rate was 0.6 m L · min-1. The column temperature was room temperature and the injection volume was 10 μL. The positive ion mode was detected by electrospray ionization and the selective reaction detection mode (SRM) was used for quantitative determination. Results: Arbutin had a good linear relationship with the peak area in the range of 1.0-150 μg · m L-1, and the recoveries of low, medium and high levels were 98.8%, 98.8% and 99.7%, respectively. The RSDs were less than 1.1%. Conclusion: The method is simple, accurate and reproducible. It can be used for the determination and quality control of arbutin in Selaginella.
