利用电子克隆技术从低温诱导的巴西橡胶树全长cDNA文库中获得了HbRD22基因,该基因全长1 458 bp。生物信息学分析结果表明,HbRD22编码一个由338个氨基酸组成的、相对分子量为36.96 ku、等电点为7.65的多肽,并含有一个由217个氨基酸组成的BURP结构域。其推导的氨基酸序列与葡萄的同源性最高,达69%。信号肽预测结果表明HbRD22蛋白为分泌型蛋白,前21个氨基酸区域为信号肽区域。Northern blotting分析结果表明,该基因在正常生长情况下,表达水平较高,干旱胁迫后24 h,表达量逐渐减低,直至检测不到,揭示该基因受干旱胁迫负调控。
The HbRD22 gene was obtained from the cryopreserved Hevea brasiliensis cDNA library by using electronic cloning technique. The full-length HbRD22 gene was 1 458 bp in length. Bioinformatics analysis showed that HbRD22 encodes a 338 amino acid peptide with a relative molecular mass of 36.96 ku and an isoelectric point of 7.65 and contains a 217 amino acid BURP domain. Its deduced amino acid sequence has the highest homology with grapes, reaching 69%. The results of signal peptide prediction indicated that the HbRD22 protein is a secreted protein and the first 21 amino acid regions are signal peptide regions. The results of Northern blotting showed that the gene was highly expressed under normal growth conditions, and the expression was decreased 24 h after drought stress until undetectable, indicating that the gene was negatively regulated by drought stress.
