目的:探讨Tiam 1基因过表达对头颈部鳞状细胞癌(HNSCC)细胞增殖及迁移能力的影响。方法:采用荧光定量RT-PCR方法检测Tiam 1基因在HNSCC细胞株中的表达;利用慢病毒介导Tiam 1基因在人HNSCC细胞株UM-SCC-47中过表达,G418筛选抗性克隆,荧光定量RT-PCR和Western blot法鉴定转导效果。利用细胞计数和四甲基偶氮唑盐法检测Tiam 1对细胞增殖的影响,划痕实验检测Tiam 1对HNSCC细胞侵袭转移的影响。结果:Tiam 1基因在高转移性HNSCC细胞株M2中呈高表达,在低转移性细胞株UM-SCC-47中呈低表达。细胞计数和MTT结果显示,过表达Tiam 1后能显著促进细胞增殖(P<0.05),划痕实验证明Tiam 1可显著促进HNSCC细胞迁移(P<0.05)。结论:Tiam 1基因具有促进HNSCC细胞增殖与迁移的能力,Tiam 1基因表达可作为HNSCC侵袭转移过程中一个有价值的指标。
Objective: To investigate the effect of Tiam 1 gene overexpression on proliferation and migration of head and neck squamous cell carcinoma (HNSCC) cells. Methods: The expression of Tiam 1 gene in HNSCC cell line was detected by fluorescence quantitative RT-PCR. The lentivirus-mediated Tiam 1 gene was overexpressed in human HNSCC cell line UM-SCC-47. The resistant clone was screened by G418, Quantitative RT-PCR and Western blot were used to identify the transduction effect. The effect of Tiam 1 on the proliferation of HNSCC cells was detected by cell counting and MTT assay. The scratch test was used to detect the effect of Tiam 1 on the invasion and metastasis of HNSCC. Results: Tiam 1 gene was highly expressed in highly metastatic HNSCC cell line M2 and low in metastatic UM-SCC-47 cell line. Cell counting and MTT results showed that Tiam 1 overexpression significantly promoted cell proliferation (P <0.05). Scratch experiments showed that Tiam 1 could significantly promote the migration of HNSCC cells (P <0.05). CONCLUSION: The Tiam 1 gene has the ability to promote the proliferation and migration of HNSCC cells. The expression of Tiam 1 gene may be a valuable indicator in the invasion and metastasis of HNSCC.
