为了筛选对多重耐药铜绿假单胞菌具有抑菌作用的中草药,并筛选铜绿假单胞菌的群体感应蛋白LasR的抑制剂,本实验利用纸片扩散法对59株临床分离的铜绿假单胞菌和1株PAO1进行抗生素药敏试验,并对其中3株菌进行中草药提取物体外抑菌试验,通过检测报告质粒中β-半乳糖苷酶的活性来筛选LasR蛋白抑制剂。实验发现,丁香、赤芍等中药粗提物具有良好的抑菌效果,部分中草药可通过影响LasR蛋白的水溶性或信号分子与受体蛋白的结合来实现对细菌群体感应的调控。
In order to screen Chinese herbal medicines with antibacterial activity against multidrug-resistant Pseudomonas aeruginosa and screen the Lasp inhibitor of Pseudomonas aeruginosa, this paper uses the disk diffusion method to detect 59 clinical isolates of Pseudomonas aeruginosa And 1 strain of PAO1 for antibiotic susceptibility testing. Three strains of bacteria were tested for antibacterial activity of Chinese herbal medicine in vitro and the LasR protein inhibitor was screened by detecting the activity of β-galactosidase in the reporter plasmid. Experiments found that clove, red peony and other crude extracts of Chinese medicine has a good antibacterial effect, part of the Chinese herbal medicine by affecting the water-soluble LasR protein or signal molecules and receptor protein binding to achieve bacterial population induction regulation.
