目的:建立大鼠血浆中橙皮苷的UHPLC-MS/MS分析测定方法,用于研究陈皮汤中橙皮苷大鼠体内的药动学。方法:采用健康大鼠灌胃给予陈皮汤,收集不同时间的含药血浆。色谱柱:BEH-C18(100 mm×2.1 mm,1.7μm);流动相为0.05%甲酸水-乙腈梯度洗脱;流速为0.3 mL·min-1;柱温35℃;进样量为5μL。结果:采用DAS 2.0软件对数据进行处理,得到大鼠血浆中橙皮苷高、中、低3个剂量组的主要药动学参数分别如下:AUC(0-t)(2 672.13±464.35)、(2 002.24±342.64)、(1 498.35±250.63)ng·h·mL-1;AUC(0-∞)(2 765.52±525.14)、(1 962.52±452.72)、(1 502.42±257.63)ng·h·mL-1;MRT(2.69±0.72)、(2.71±0.57)、(2.65±0.21)h;T1/2(1.39±0.15)、(1.42±0.08)、(1.51±0.14)h;Cmax(1 124.63±356.62)、(914.35±244.52)、(742.24±51.25)ng·mL-1;Tmax(1.45±0.03)、(1.46±0.02)、(1.51±0.01)h。结论:实验结果表明该方法特异性好、速度快。灵敏度高等优点,可用以橙皮苷大鼠体内药动学研究。
OBJECTIVE: To establish a method for the determination of hesperidin in rat plasma by UHPLC-MS / MS method and to study the pharmacokinetics of hesperidin in Fructus Mumex. Methods: The healthy rats were given gavage soup, collected at different times of the drug-containing plasma. Column: BEH-C18 (100 mm × 2.1 mm, 1.7 μm). The mobile phase was eluted with 0.05% formic acid in water and acetonitrile gradient. The flow rate was 0.3 mL · min-1. The column temperature was 35 ℃ and the injection volume was 5 μL. Results: The main pharmacokinetic parameters of high, middle and low dose of hesperidin in rats plasma were obtained by DAS 2.0 software. The main pharmacokinetic parameters were as follows: AUC (0-t) (2672.13 ± 464.35), (2 002.24 ± 342.64), (1 498.35 ± 250.63) ng · h · mL-1, AUC (0-∞) (2 765.52 ± 525.14), (1 962.52 ± 452.72) and (1 502.42 ± 257.63) ng · h · ML -1; MRT (2.69 ± 0.72), (2.71 ± 0.57) and (2.65 ± 0.21) h respectively; T1 / 2 (1.39 ± 0.15), (1.42 ± 0.08) and 124.63 ± 356.62, (914.35 ± 244.52), (742.24 ± 51.25) ng · mL-1; Tmax (1.45 ± 0.03), (1.46 ± 0.02) and (1.51 ± 0.01) h. Conclusion: The experimental results show that the method is specific and fast. High sensitivity and other advantages, can be used hesperidin rat pharmacokinetic studies.
