目的 比较SUS-MDCK、MDCK和SPF鸡胚3种方法对流感样病例(influenza-likeillness,ILI)标本中不同型别流感病毒的分离培养敏感性差异,初步评估SUS-MDCK在流感监测中的应用前景和改进方向.方法 选取2018年10月至2019年6月沈阳市流感监测哨点医院采集的ILl标本中流感病毒核酸阳性的标本,同时应用SUS-MDCK、MDCK和SPF鸡胚3种方法进行病毒分离,对分离结果资料进行比较分析.结果 1 296份ILI标本中,核酸阳性率为15.35%(199/1 296);毒株分离情况为:SUS-MDCK细胞毒株172株、MDCK细胞毒株160株、鸡胚株54株.结论 对于季节性流感病毒的分离培养,细胞法优于鸡胚法;SUS-MDCK和MDCK在分离人季节性流感临床标本的敏感性上,因毒株型别不同而略有差异,H3N2型前者好于后者,甲型H1N1型两者相当,而B型victoria系后者略优于前者.从病毒滴度来看,MDCK细胞毒株和SPF鸡胚毒株相当,但SUS-MDCK细胞毒株高于前两者.“,”Objective To compare the sensitivities of the three culture methods using suspension MDCK (SUS-MDCK) cells,adherent MDCK cells and embryonated chicken egg in isolating different types of influenza virus from clinical specimens of influenza like illness (ILI) cases,so as to evaluate the trend on application and improvement of SUS-MDCK in influenza surveillance.Methods The ILl samples with positive results of influenza virus nucleic acids were selected from the samples collected in the sentinel hospitals in Shenyang from October 2018 to June 2019.The three culture methods with SUSMDCK cells,adherent MDCK cells and embryonated chicken egg were performed in parallel to isolate the virus and the data of virus isolation were analyzed.Results For 1 296 ILI samples,the rate of nucleic acid positive was 15.35% (199/1 296).The numbers of influenza virus strains isolated by different culture methods were 172 by SUS-MDCK cells,160 by adherent MDCK cells and 54 by embryonated chicken egg.Conclusions The cell culture methods were better for seasonal influenza virus isolation than the culture with embryonated chicken egg.The sensitivities of isolating different types of human seasonal influenza virus by SUS-MDCK cells and adherent MDCK cells were slightly different.The SUS-MDCK cells was better than adherent MDCK cells in the isolation of influenza A (H3N2) strains while both were suitable for influenza A (H1NI) strains.However,isolation of influenza B/Victoria-Lineage virus by adherent MDCK cells was slight better than SUS-MDCK cells.The titers of virus isolated by adherent MDCK cells and embryonated chicken egg were equal while the titer by SUS-MDCK cell was higher than both methods above.
