用EB病毒转化微量全血法建立了一对具高SCE特征的习惯流产夫妇的B淋巴母细胞株:JMSW-1,JMSW-2。细胞株平均倍增时间分别为45.5小时/代;48小时/代。常规染色体核型分析为正常核型且均保持高SCE特征。JMSW-1自发SCE为14.00次/每细胞,JMSW-2自发SCE为13.28次/每细胞。微核率分别为24.5‰;,20.5‰,加0.01μg/ml丝裂霉素C(MMC)处理,诱发SCE及微核率与正常细胞株相比均显著升高。JMSW-1诱发SCE为23.58次/每细胞,微核率42‰;JMSW-2诱发SCE为22.72次/每细胞,微核率39.5%;,夫妇双方自发、诱发SCE均高于正常对照组(P<0.01)。
A B-lymphoblastoid cell line was established with a couple of conventional abortion couples with high SCE characteristics using Epstein-Barr virus transformation micro-whole blood method: JMSW-1, JMSW-2. The average cell doubling time were 45.5 hours / generation; 48 hours / generation. Conventional karyotype analysis was normal karyotype and maintained high SCE characteristics. The spontaneous SCE of JMSW-1 was 14.00 cells / cell, and the spontaneous SCE of JMSW-2 was 13.28 cells / cell. The micronuclei rates were 24.5 ‰ and 20.5 ‰, respectively. The addition of 0.01 μg / ml MMC increased the SCE and micronucleus rates significantly compared with those of normal cell lines. JMSW-1 induced SCE was 23.58 times per cell, micronucleus rate was 42 ‰; JMSW-2 induced SCE was 22.72 times per cell, micronucleus rate was 39.5%; both spontaneous and induced SCE Higher than the normal control group (P <0.01).
