目的 :研究抗小鼠胸腺基质细胞 (MTSC)Pf18 3单抗识别分子的表达特性。方法 :采用流式细胞 (FACS)和激光扫描共焦显微镜 (CLSM)分析TSC单抗Pf18 3染色特征。结果 :FACS分析发现Pf18 3单抗识别分子可表达于胸腺、胎肝和骨髓等不同来源的基质细胞系和腹腔巨噬细胞表面。新鲜分离的胸腺细胞Pf18 3识别分子表达阴性 ,但ConA活化可诱导并上调其表达 ,随活化时间延长表达增高 ,且主要表达于CD4+CD8+和CD4+CD8-两细胞亚群。CLSM分析可见 :荧光沿胸腺细胞边缘呈环状分布 ,活化后荧光明显增厚向胞浆延伸 ,胞核不着色。结论 :Pf18 3单抗识别分子共表达于胸腺基质细胞及活化的胸腺细胞 ,并与CD4+CD8+和CD4+CD8-亚群胸腺细胞活化密切相关
AIM: To investigate the expression characteristics of anti-mouse MTS Pf18 3 monoclonal antibody. Methods: The characteristics of Pf18 3 staining of TSC McAb were analyzed by flow cytometry (FACS) and confocal laser scanning microscopy (CLSM). Results: FACS analysis showed that Pf18 3 mAb was expressed on stromal cell lines and peritoneal macrophages from different sources, such as thymus, fetal liver and bone marrow. Freshly isolated thymocytes were negative for Pf18 3, but ConA activation induced and up-regulated the expression of Pf18 3, increased with activation time and mainly expressed in both CD4 + CD8 + and CD4 + CD8- cell subsets. CLSM analysis shows: Fluorescent distribution along the thymus edge of the ring, significantly increased after activation of fluorescence to the cytoplasm extension, the nucleus is not colored. CONCLUSION: Pf18 3 mAb is co-expressed in thymocytes and activated thymocytes, and is closely related to the activation of thymocytes in CD4 + CD8 + and CD4 + CD8- subsets
