目的:观察复方柔肝抑纤饮抗肝纤维化的疗效,并探讨其机制。方法:制备大鼠实验性肝纤维化模型,进行相应分组给予不同的处理,考察血清肝功能和肝纤维化指标,并对肝脏组织行免疫组化染色、Masson染色,结合图像定量分析等技术分析Ⅰ型胶原(col-Ⅰ)、Ⅲ型胶原(col-Ⅲ)水平。结果:柔肝抑纤饮可明显改善大鼠肝纤维化,并使血清丙氨酸氨基转移酶(ALT)、透明质酸(HA)等指标降低,降低col-Ⅰ、col-Ⅲ水平,经统计分析有显著性差异。结论:柔肝抑纤饮具有抗脂质过氧化作用,降低了col-Ⅰ、col-Ⅲ的抑制表达,可明显减轻大鼠肝脏内胶原纤维增生沉积,减少纤维化组织面积,减少了肝脏细胞外基质的产生和沉积,从而达到抗肝纤维化的作用。
Objective: To observe the curative effect of compound Rougan Yixian Decoction on hepatic fibrosis and its mechanism. Methods: Experimental rat model of hepatic fibrosis was established. The rats were divided into different groups for different treatment. The serum liver function and liver fibrosis indexes were examined. The liver tissues were analyzed by immunohistochemistry, Masson staining, combined with image quantitative analysis Col-Ⅰ and Col-Ⅲ. Results: Rougan Yixian Decoction can significantly improve hepatic fibrosis in rats and decrease serum ALT, HA and other indexes, and decrease the levels of col-Ⅰ and col-Ⅲ Statistical analysis showed significant differences. Conclusion: Rougan Yixian Decoction can inhibit the expression of col-Ⅰ and col-Ⅲ, reduce the proliferation of collagen fibers in the liver, reduce the area of fibrosis tissue and decrease the number of liver cells External matrix generation and deposition, so as to achieve the role of anti-liver fibrosis.
