目的 比较角膜与三叉神经节 (TG)内单纯疱疹病毒Ⅰ型 (HSV 1)潜伏相关转录 (LAT)启动子序列 ,进一步证实角膜为HSV 1的另一潜伏地。方法 用新西兰白兔建立HSV 1潜伏感染动物模型 ,提取并PCR扩增角膜及TGLAT启动子 ,比较LAT启动子序列。结果 6 6 7% (4/6 )的角膜组织扩增出HSV 1LAT启动子 ,且与TG的电泳位置相同 (2 16bp) ;其在 2~ 16 1间的核苷酸序列完全相同。结论 HSV 1潜伏期的角膜组织存在LAT启动子 ,并与TG内的序列相同 ,表明角膜在很大程度上与TG一样 ,是HSV 1的另一潜伏点。
Objective To compare the latent transcriptional (LAT) promoter sequence of herpes simplex virus type 1 (HSV 1) in the cornea and trigeminal ganglion (TG) to further confirm that the cornea is another latent site of HSV 1. Methods Animal models of latent infection with HSV 1 were established in New Zealand white rabbits. The cornea and TGLAT promoter were extracted and amplified. The LAT promoter sequence was compared. Results 6 6 7% (4/6) of corneal tissue amplified HSV 1 LAT promoter, and the electrophoresis position of TG was the same (2 16 bp); its nucleotide sequence was exactly the same between 2 and 16 1. Conclusions The presence of the LAT promoter in the HSV 1 latent period of corneal tissue and the same sequence as in the TG indicate that the cornea is largely the same as TG and is another lurking point for HSV 1.
