二甲基黄(DAB)诱发大白鼠肝癌癌变过程中食物因素对鼠肝核酸、磷脂的含量有影响,已见前述。这可能是由于肝癌癌变过程中肝细胞核膜的通透性改变、基因表达的调控机理异常,和混合功能氧化酶系统的功能改变,文献也已陆续述及。 为了进一步探讨DAB诱发肝癌的大白鼠肝脏核酸的变化,我们建立了肝细胞核与肝细胞质的分步分离,核酸的提取与纯化,核糖核酸(RNA)体外同位素碘标记及固相核酸分子杂交等技术,并作了正常鼠与DAB诱发肝癌的鼠肝核酸杂交竞争抑制试验,现报告如下。
