补阳还五汤对中风后遗症“气虚血瘀”大鼠模型血管内皮生长因子表达的影响

目的:探讨中风后遗症“气虚血瘀”大鼠模型血管内皮生长因子(VEGF)的表达及补阳还五汤对其表达的影响。方法:采用机械开颅电凝法阻断大脑中动脉24h后,用随机电脉冲刺激仪每天刺激大鼠2h,连续刺激15天,复制中风后遗症“气虚血瘀”大鼠模型。治疗组在造模结束后灌服补阳还五汤水煎液,连续给药15天。通过重量法观察脑含水量的变化,并采用免疫组织化学方法和酶联免疫方法检测VEGF的表达和蛋白水平。结果:与模型组相比,补阳还五汤能降低模型大鼠脑含水量(P<0.05),VEGF阳性细胞数量明显增多(P<0.05),蛋白水平提高(P<0.05)。结论:补阳还五汤能改善中风后遗症大鼠脑水肿,并增强VEGF的表达和蛋白水平,这可能是其发挥抗脑缺血损伤作用的机制之一。 Objective: To investigate the expression of vascular endothelial growth factor (VEGF) in the rat model of apoplexy “qi deficiency and blood stasis” and the effect of Buyang Huanwu Decoction on its expression. METHODS: After the middle cerebral artery was blocked with mechanical craniotomy for 24 hours, the rats were stimulated with a random electrical pulse stimulation instrument for 2 hours per day for 15 consecutive days. The rat model of apoplexy syndrome with Qi deficiency and blood stasis was reproduced. The treatment group was fed with decoction of Buyang Huanwu Decoction at the end of the model and continued for 15 days. The change of brain water content was observed by gravimetric method, and the expression of VEGF and protein levels were detected by immunohistochemical method and enzyme-linked immunosorbent assay. Results: Compared with the model group, Buyang Huanwu Decoction can reduce the brain water content (P<0.05), increase the number of VEGF-positive cells (P<0.05), and increase the protein level (P<0.05). Conclusion: Buyang Huanwu Decoction can improve cerebral edema in rats with stroke sequelae, and increase the expression of VEGF and protein, which may be one of the mechanisms of anti-cerebral ischemic injury.
