目的:观察清洁级雄性SD大鼠五点注射卵蛋白及臭氧暴露后肺功能、肺组织病理HE染色以及血清氧化应激指标表现,初步探索氧化应激特征性支气管哮喘SD大鼠模型的建立方法;方法:30只雄性SD大鼠随机分为3组,即空白对照A组、卵蛋白B组和卵蛋白复加臭氧C组。B、C组大鼠五点注射10%卵蛋白(OVA),A组大鼠五点注射等剂量生理盐水,每周1次,共两次;C组大鼠注射OVA后隔天进行臭氧暴露;14 d后B、C组用1%OVA连续雾化7 d诱发其哮喘发作,C组雾化同时仍隔天进行臭氧暴露,A组则给予生理盐水雾化吸入。结果:(1)B、C两组肺功能结果较A组差异有统计学意义,臭氧暴露对哮喘大鼠气道重塑有一定影响;(2)HE染色可见:B、C组支气管可见炎性细胞浸润,上皮细胞脱落;肺泡塌陷,融合;C组相比炎性浸润更为密集;(3)B、C两组血清对氧自由基的清除能力降低,与A组比较差异有统计学意义;臭氧暴露建模影响大鼠的抗氧化能力;结论:SD大鼠五点注射卵蛋白并复加臭氧暴露可成功建立氧化应激特征性支气管哮喘模型。
OBJECTIVE: To observe the five-point injection of ovalbumin in clean male SD rats and lung function after exposure to ozone, the histopathological HE staining and the serum oxidative stress indexes in SD rats, and to explore the establishment of a rat model of oxidative stress-induced bronchial asthma Methods: Thirty male Sprague-Dawley rats were randomly divided into three groups: blank control group A, ovalbumin B group and ovalbumin plus ozone C group. Rats in group B and C were injected orally with 10% ovalbumin (OVA) at 5 o’clock. Rats in group A were injected with equal doses of normal saline at 5 o’clock once a week for two times. Rats in group C received ozonation After 14 days, B and C groups were challenged with 1% OVA for 7 days to induce asthma attacks. In group C, ozone exposure was also performed on the next day while group A was given saline inhalation. Results: (1) The results of lung function in group B and group C were significantly different from those in group A, and the ozone exposure had a certain influence on airway remodeling in asthmatic rats. (2) The results of HE staining showed that the bronchitis of group B and C was visible (3) The ability of scavenging oxygen free radicals in serum of groups B and C was lower than that of group A, and the difference was statistically significant compared with that of group A Significance; Ozone exposure modeling can affect the antioxidant capacity of rats. CONCLUSION: Five-point injection of ovalbumin and ozone exposure in SD rats successfully established a model of oxidative stress-induced bronchial asthma.
