目的研究实验性肝纤维化时碱性成纤维细胞生长因子(bFGF)在肝脏的表达.方法实验大鼠分肝硬变组(24只)和正常对照组(8只),肝硬变组采用复合因子制作出肝硬变动物病理模型,通过免疫组化研究了bFGF在肝纤维化过程中的表达.同时还通过结蛋白(Desmin,DM),ED1的免疫组化变化,反映肝纤维化过程中储脂细胞和肝枯否细胞增生情况.结果随着肝纤维化发展,DM,ED1和bFGF阳性细胞在肝窦,坏死区和纤维间隔逐渐增多,bFGF阳性细胞主要是在那些DM阳性细胞和ED1阳性细胞中表达,定量分析,各组均高于正常组,以6周组最高,ED1阳性细胞与bFGF和DM阳性细胞的变化成正相关关系(r=0977和r=0989,P<005),bFGF阳性细胞和DM阳性细胞的变化也成正相关关系(r=0997,P<005).结论在肝纤维化发生时由于肝枯否细胞和储脂细胞的增生及二者表达bFGF增加,再通过bFGF的自分泌和旁分泌作用进一步造成储脂细胞的增生和ECM的产生增加.
Objective To investigate the expression of basic fibroblast growth factor (bFGF) in liver during experimental liver fibrosis. Methods The experimental rats were divided into cirrhosis group (n = 24) and normal control group (n = 8). The liver cirrhosis group was made with compound factor to make animal model of cirrhosis. Immunohistochemistry was used to study the effect of bFGF on hepatic fibrosis In the expression. At the same time, the expression of Desmin (DM) and ED1 was also examined by immunohistochemistry to show the proliferation of fat-storing cells and hepatic cells in liver fibrosis. Results With the development of hepatic fibrosis, DM, ED1 and bFGF-positive cells gradually increased in the sinusoidal, necrotic and fibrous septum. The bFGF-positive cells were mainly expressed in DM-positive cells and ED1-positive cells, and were quantitatively analyzed. The positive rate of ED1 positive cells was positively correlated with the changes of bFGF and DM positive cells (r = 0977 and r = 0989, P <005). The positive rate of bFGF positive cells and DM There was also a positive correlation between the changes of positive cells (r = 0997, P <005). Conclusions The hepatic fibrosis results in the proliferation of lipid-storing cells and the increase of ECM due to the proliferation of hepatic fibrosis cells and fat-storing cells and the increase of both bFGF and bFGF through autocrine and paracrine effects.
