以新疆恰玛古种子萌发5d无菌苗的子叶及下胚轴为外植体,研究了各种不同激素配比条件下新疆恰玛古的再生体系。结果表明:培养基MS+2,4-D 2.0mg/L+KT 0.1mg/L对愈伤组织诱导效果较好,子叶、下胚轴的愈伤诱导率分别为65.4%、95.5%;再生芽的诱导培养基为6-BA 3.0mg/L+NAA 0.5mg/L+AgNO35.0mg/L,子叶、下胚轴的芽分化率分别为20.8%、65.0%。将芽转入生根培养基MS+IBA 0.5mg/L中诱导生根。20d后,生根的小植株练苗移入花土中,成活率达95%。
The cotyledons and hypocotyls of germ-free 5-day germ-free seedlings of Xinjiang Qiama ancient seed were used as explants to study the regeneration system of Qiamagu in Xinjiang under different hormone combinations. The results showed that the medium of MS + 2,4-D 2.0 mg / L + KT 0.1 mg / L had better effect on callus induction, and the callus induction rates of cotyledon and hypocotyls were 65.4% and 95.5% respectively. The bud induction medium was 6-BA 3.0 mg / L + NAA 0.5 mg / L + AgNO 35.0 mg / L, and the bud differentiation rates of cotyledons and hypocotyls were 20.8% and 65.0%, respectively. The shoots were transferred to rooting medium MS + IBA 0.5mg / L to induce rooting. After 20 days, the rooting plantlets were moved into flower soil, the survival rate reached 95%.
