本文介绍用高效液相色谱内标准法同时测定人血浆中视黄醇(维生素A)和α-及γ-生育酚(维生素E)。用美国Beckman344系列高效液相色谱仪,165型可调紫外一可见分光检测器及日本岛津C—RIB微处理机。分析柱为C_(13)反相柱,前加一短予柱。流动相为甲醇:水(98:2)。紫外波长300nm下检测。色谱分析于12分钟内完成。用内标准二点计算法,从峰面积比值计算结果,以免因标准曲线不通过零点所引起的计算误差。回收率的测定结果,视黄醇之平均回收率为93.9%,α-生育酚为106.9%。变异系数的结果,视黄醇为
This article describes the simultaneous determination of retinol (vitamin A) and α- and γ-tocopherol (vitamin E) in human plasma by HPLC-internal standard method. With the United States Beckman344 series of high performance liquid chromatography, 165 adjustable UV-visible spectrophotometric detector and Japan Shimadzu C-RIB microprocessor. The analytical column was C_ (13) reversed phase column, followed by a short pre-column. The mobile phase was methanol: water (98: 2). UV wavelength 300nm detection. Chromatography was completed in 12 minutes. With the internal standard two-point calculation method, the results from the peak area ratio calculation, so as to avoid the standard curve does not pass zero calculation error caused.
