目的:建立钩葛定眩颗粒的质量标准。方法:采用薄层色谱法鉴别钩藤、川芎和丹参;采用高效液相色谱法测定葛根素、丹酚酸B的含量。结果:薄层色谱斑点清晰,分离度好;高效液相色谱法测定葛根素、丹酚酸B的含量,葛根素在5.015~80.240μg/mL的浓度范围内线性关系良好(r=0.999 8,n=6),平均回收率为99.14%,RSD为1.47%(n=9);丹酚酸B在10.04~160.64μg/mL范围内线性关系良好(r=0.999 7,n=6),平均回收率为98.86%,RSD为1.58%(n=9)。结论:建立的定性定量方法操作方便、灵敏度高、结果准确、专属性强,可用于钩葛定眩颗粒的质量控制。
OBJECTIVE: To establish the quality standard of HAGETDING dizzy granules. Methods: Uncaria rhizoma, Rhizoma Chuanxiong and Salvia miltiorrhiza were identified by TLC. The content of puerarin and salvianolic acid B was determined by high performance liquid chromatography. Results: The TLC spots were clear and the resolution was good. The content of puerarin and salvianolic acid B was determined by HPLC. The calibration curve was linear in the range of 5.015 ~ 80.240μg / mL (r = 0.999 8, n = 6). The average recovery was 99.14% and the RSD was 1.47% (n = 9). The linear range of salvianolic acid B in the range of 10.04-160.64μg / mL was good (r = 0.999 7, n = 6) The recovery was 98.86% with a RSD of 1.58% (n = 9). Conclusion: The established qualitative and quantitative method is easy to operate with high sensitivity, accurate results and high specificity. It can be used for the quality control of HGGD.
