目的 观察 1,2 5 二羟基维生素D3 [1,2 5 (OH) 2 D3 ]对白血病细胞株HL 6 0的诱导分化作用并探讨其机制。方法 通过MTT试验、细胞形态观察、表面标志分析和硝基四氮唑蓝 (NBT)还原反应 ,观察 1,2 5 (OH) 2 D3 作用后HL 6 0细胞增殖的改变和分化情况 ;通过流式细胞仪和RT PCR检测细胞周期和nm2 3基因表达的变化。结果 HL 6 0细胞经 1,2 5 (OH) 2 D3 作用后增殖受抑制、向成熟单核细胞系分化、细胞被阻滞在G1 期、nm2 3基因的表达下调。结论 1,2 5 (OH) 2 D3 对HL 6 0细胞株具有诱导分化作用 ,其作用机制可能与下调nm2 3基因的表达有关。
Objective To observe the effect of 1,25-dihydroxyvitamin D3 [1,25(OH) 2 D3] on the differentiation of leukemia cell line HL 60 and investigate its mechanism. Methods MTT assay, cell morphology, surface marker analysis and nitroblue tetrazolium (NBT) reduction were used to observe the proliferation and differentiation of HL 60 cells after 1,25(OH) 2 D3 treatment. Cytometry and RT PCR were used to detect changes in cell cycle and nm23 gene expression. Results The proliferation of HL-60 cells was inhibited by 1,25 (OH) 2 D3, and they were differentiated into mature monocytes. The cells were arrested in G1 phase and the expression of nm23 gene was down-regulated. Conclusion 1,25(OH) 2 D3 can induce differentiation of HL 60 cell line, and its mechanism may be related to the down-regulation of nm23 gene expression.
