目的副流感病毒兰州株的分离与鉴定。方法应用鸡胚和人胚肺成纤维突变细胞株分离病毒 ;应用血凝和血抑以及近年来多株流感国际和国家株 (H1N1亚型 6株、H3N2亚型 2株、B型 4株 )作抗原抗体交互试验 ;还用 4类主要呼吸道病毒单克隆抗体间接免疫荧光法与 2株兰州株的病毒抗原基质片作特异性交叉反应。结果 2株兰州株在鸡胚中培养血凝效价滴度高达 1:5 12 (+ + )。它们与流感病毒甲型 (H1N1和 H3N2 )以及乙型流感抗血清均无血抑反应。它们的血凝素抗原及血清与 12株甲和乙型流感病毒株无免疫血抑交互反应。但 2株流行株抗原片可和副流感病毒 2型单克隆抗体发生较强反应 ,在人胚肺细胞中培养有明显细胞融合病变。结论 2株流行株虽具有高滴度血凝效价 ,但不是流感病毒株 ,根据单克隆抗体荧光染色和流行株在人胚肺细胞中的病变 ,初步确认为副流感病毒 2型
Isolation and identification of the parainfluenza virus Lanzhou strain. Methods The virus was isolated from chick embryo and human embryo lung fibroblast cell lines. Hemagglutination and hemopoiesis as well as international and national strains (H1N1 subtype, 2 H3N2 subtype and 4 B type) As antigen-antibody interaction test; also four kinds of major respiratory virus monoclonal antibody indirect immunofluorescence method with two strains of Lanzhou virus antigen matrix specific cross-reaction. Results The titer of hemagglutination titer of two strains of Lanzhou was up to 1: 5 12 (+ +) in chicken embryos. They have no hematocrit response to the influenza A (H1N1 and H3N2) and influenza B antisera. Their hemagglutinin antigen and serum had no immunosuppressive interaction with 12 strains of influenza A and B viruses. However, two strains of epidemic strains can react strongly with the parainfluenza virus type 2 monoclonal antibody and have obvious cell fusion lesions in human embryo lung cells. Conclusions Although the two strains have high titers of titer and hemagglutination titer, they are not influenza virus strains. According to the monoclonal antibody fluorescent staining and the epidemic strains in human embryo lung cells, the two strains were initially identified as parainfluenza 2
