FraXA定位于Xq27,是一罕见型脆性位点,只有X连锁智能障碍患者在胸苷酸缺乏的培养条件下表达。但本文作者曾在两个正常男性和一只雄性黑猩猩中发现在同样的培养条件下其Xq27处有4~5%的脆性表达,故而作者推测Xq27处有一普通型脆性位点,即c-fra(X)(927)的存在。为了确证这一点,作者在本文进一步研究了四个脆性X男性、七个无遗传病家族史的正常男性、两只正常的雄性黑猩猩。标本细胞在RPMI1640+10%胎牛血清中进行培养,于收获前24小时进行①10~(-8)M FudR
FraXA is located at Xq27, a rare type of fragile site that is only expressed in cultures with thymidylate deficiency in X-linked mental retardation. However, the authors found that in two normal males and one male chimpanzee, there is a 4-5% friability expression at Xq27 under the same culture conditions. Therefore, the authors speculate that there is a common type of brittle site at Xq27, namely c-fra (X) (927). To confirm this point, the authors further investigate four fragile X males, seven normal males with a family history of no genetic disease, and two normal male chimpanzees in this article. The specimen cells were cultured in RPMI1640 + 10% fetal bovine serum and subjected to ①10 ~ (-8) M FudR 24 hours before harvest
