目的 从肺癌患者外周血诱导扩增的树突状细胞 ,观察其形态、表型、抗原递呈功能和对LAK细胞抗肿瘤作用的影响。方法 肺癌患者外周血单个核细胞经GM -CSF、IL - 4诱导扩增成为树突状细胞 ,FACS检测其表型 ,MLR测定其抗原递呈能力及对LAK细胞增殖的影响。TAA冲击的DC与LAK细胞混合培养诱导的CTL ,用MTT法测定其细胞毒。结果 肺癌患乾外周血诱导的树突状细胞具有典型的形态、表型特征和激发MLR的能力 ,TAA冲击的DC能显著地提高LAK细胞的增殖能力和增强对靶抗原细胞的细胞毒效应。结论 从肺癌患者外周血中可获得典型的树突状细胞 ,经TAA冲击的DC能从LAK细胞中诱导出特异性CTL细胞 ,以该DC为基础的疫苗联合LAK细胞 ,可能在肺癌生物治疗中发挥重要的作用
OBJECTIVE: To induce dendritic cells (DCs) from peripheral blood of patients with lung cancer to observe their morphology, phenotype, antigen presenting function and anti-tumor effect on LAK cells. Methods Peripheral blood mononuclear cells (PBMCs) of lung cancer patients were induced to proliferate into dendritic cells by GM-CSF and IL-4. The phenotypes of the dendritic cells were detected by FACS. The antigen presenting ability and the effect on the proliferation of LAK cells were determined by MLR. TAA-challenged DCs were mixed with LAK cells to induce induced CTLs and their cytotoxicity was measured by MTT assay. Results The dendritic cells induced by peripheral blood of patients with lung cancer had typical morphology, phenotypic characteristics and the ability to stimulate MLR. TAA shocked DC significantly increased the proliferation of LAK cells and enhanced the cytotoxic effect on target antigens. Conclusion Typical dendritic cells can be obtained from the peripheral blood of patients with lung cancer. TAA-challenged DCs can induce specific CTL cells from LAK cells. Combination of this DC-based vaccine with LAK cells may be involved in the biological treatment of lung cancer Play an important role
