目的 研究Kir2 1和Kir3 1mRNA在平滑肌及其它组织中表达的差异。方法 用RT PCR方法检测mRNA的表达 ,用光密度扫描法半定量测定表达差异。结果 在大鼠的左心室、大脑皮层、主动脉平滑肌、膀胱平滑肌和呼吸道平滑肌上均有Kir2 1和Kir3 1的表达 ,通过光密度扫描分析显示Kir2 1mRNA在各组织的表达水平 (Kir2 1/GAPDH)依次为主动脉平滑肌 >心脏 =大脑皮层 =气管平滑肌 >膀胱平滑肌 ;Kir3 1mRNA的表达水平 (Kir3 1/GAPDH)依次为主动脉平滑肌 =气管平滑肌 >心脏 =大脑皮层 =膀胱平滑肌。结论 Kir2 1和Kir3 1在不同组织中2 0 0 0 0 4 0 5收稿 国家自然科学基金资助项目 ,No 39770 85 9;国家重点基础研究“973”子课题 ,NoG19980 5 110 6作者简介 :任亚军 ,男 ,30岁 ,博士研究生 ;王晓良 ,男 ,45岁 ,PhD (德国 ) ,研究员 ,博士生导师 ,中国药理学会常务理事的表达量是有差异的 ,这种差异有内在的生理意义。
Objective To investigate the differences of Kir2 1 and Kir3 1 mRNA expressions in smooth muscle and other tissues. Methods RT PCR method was used to detect the expression of mRNA. Semiquantitative determination of the expression difference by optical density scanning. Results Kir2 1 and Kir3 1 were expressed in left ventricle, cerebral cortex, aortic smooth muscle, bladder smooth muscle and respiratory smooth muscle in rats. The expression of Kir2 1 mRNA in each tissue was detected by optical density scanning (Kir2 1 / GAPDH ) Followed by aortic smooth muscle> cardiac = cerebral cortex = tracheal smooth muscle> bladder smooth muscle; Kir3 1 mRNA expression level (Kir3 1 / GAPDH) followed by aortic smooth muscle = tracheal smooth muscle> cardiac = cerebral cortex = bladder smooth muscle.
