Objective: To establish an animal model of reduced bone mass caused by long-term increasing load exercise. Methods: Three-month-old female rats with estrous cycle regularity were randomly divided into 8 groups, and the rats were divided into 2 groups, 6, 6, 8, 9, 11, 13 and 15 weeks of incremental load exercise. In the modeling process to monitor body weight, body composition, vaginal exfoliated cells, blood biochemistry and bone mineral density and other indicators of dynamic changes. Results: During the treadmill exercise with increasing load for 15 weeks, rats gradually showed the characteristics of excessive training such as increased BUN, body weight, percentage of body fat and Hb decline. The level of E2 decreased continuously and the inhibition of estrous cycle deepened finally. BMD decreased. The results showed that the 15-week incremental load treadmill exercise successfully established an animal model of excessive exercise-induced bone loss.
