目的:观察大鼠脑梗死后脑内自体神经干细胞(Neural stem cells,NSCs)的增殖、迁移与分化以及祛痰通络中药对其影响。方法:Sprague-Dawley(SD)大鼠随机分为假手术组(N组)、生理盐水组(A组)、祛痰通络中药治疗组(B组),比较各组大鼠在脑梗死模型建立后24h、3天、7天、14天各时间点神经功能缺损评分,并应用免疫组化法检测并比较各时间点自体神经干细胞增殖情况、免疫荧光双标后观察增殖干细胞的分化情况。结果:脑梗死后第14天祛痰通络中药治疗组评分低于生理盐水组(P<0.05),第3天开始增殖细胞、第7天开始分化为神经元样细胞数均多于A组(P<0.05)。结论:祛痰通络中药可促进脑梗死后自体神经干细胞的增殖并且更多的分化为神经元,从而促进脑梗死大鼠的神经功能恢复。
OBJECTIVE: To observe the proliferation, migration and differentiation of neural stem cells (NSCs) in the brain after cerebral infarction in rats and the effect of expectorant and collaterals on it. Methods: Sprague-Dawley (SD) rats were randomly divided into sham operation group (N group), normal saline group (Group A) and expectorant Tongluo Chinese medicine treatment group (B group) Neurological deficit scores were established at 24h, 3d, 7d, 14d after operation. Proliferation of autologous neural stem cells was detected by immunohistochemistry. Differentiation of proliferating stem cells was observed by double immunofluorescence staining. Results: On the 14th day after cerebral infarction, the scores of expectorant and collaterals group were lower than that of the normal saline group (P <0.05). The number of proliferating cells began to proliferate on the third day, (P <0.05). Conclusion: Qutan Tongluo can promote the proliferation of autologous neural stem cells after cerebral infarction and further differentiate into neurons, so as to promote the recovery of neurological function in rats with cerebral infarction.
