目的:利用等位酶与可溶性蛋白凝胶电泳技术探讨远志不同种源的鉴别与亲缘关系。方法:采用聚丙烯酰胺凝胶电泳(PAGE)技术分析不同种源远志苹果酸酶、过氧化物酶、酯酶与可溶性蛋白谱带,采用NASYS软件对电泳谱带进行聚类分析。结果:远志可溶性蛋白与等位酶聚丙烯酰胺凝胶电泳图谱能够鉴别不同种源的远志,9份不同种源远志可被聚为三种类型,遗传距离0.1054~1.2040,除卵叶远志外,其余8份远志遗传距离0.1054~0.6931,卵叶远志与远志间的遗传距离明显高于远志间的遗传距离,地理位置相近的远志遗传距离较小,亲缘关系较近。结论:利用等位酶与可溶性蛋白凝胶电泳技术可为不同种源远志的鉴定与划分提供参考。
OBJECTIVE: To study the identification and genetic relationship of different provenances of Polygala tenuifolia by using allozyme and soluble protein gel electrophoresis. Methods: Polyacrylamide gel electrophoresis (PAGE) was used to analyze the bands of malic enzyme, peroxidase, esterase and soluble protein in Polygala tenuifolia from different provenances. The clustering analysis was carried out by using NASYS software. Results: Polygala tenuifolia and polyacrylamide gel electrophoresis of allozyme could identify Polygala tenuiflora from different provenances. Polygala tenuifolia could be clustered into three types with the genetic distance of 0.1054-1.2040, The remaining 8 copies of Polygalaceae genetic distance 0.1054 ~ 0.6931, Eggplant Polygalaceae and Polygalaceae between the genetic distance was significantly higher than between Polygalaceae genetic distance Polygala similar geographical proximity to a smaller genetic distance, closer genetic relationship. Conclusion: Allozyme and soluble protein gel electrophoresis can provide reference for the identification and classification of Polygala tenuiflora from different provenances.
