探讨玛咖中总皂苷含量测定的方法。以《保健食品检验与评价技术规范(2003版)》中“保健食品中总皂苷的测定”为基础,考察用D101大孔吸附树脂代替Amberlite-XAD-2大孔树脂纯化总皂苷的可行性,并研究样品在树脂中的吸附时间、样品总皂苷的浓度两方面对检测结果的影响。结果表明,标准曲线在0~0.176 mg/m L范围内线性关系良好,线性回归方程y=3.308 4x-0.006 8(R2=0.999 0),方法检出限为2μg/m L。两种树脂的总皂苷检测结果无显著差异。样品吸附时间为60 min,样品中总皂苷含量在0.08~0.32 mg/m L范围内时,总皂苷的检测误差小。改进后的方法平均加标回收率99.76%,RSD为2.23%(n=6),精密度试验RSD为2.97%(n=6)。改进后的总皂苷检测方法灵敏度高,准确性好,重现性好,可用于测定玛咖中总皂苷的含量,玛咖中总皂苷含量为3.81±0.11 mg/g(n=6)。
To explore the method of determination of total saponin content in maca. Based on “Determination of total saponins in health food” in “Technical Specification for Testing and Evaluation of Health Food (2003 Edition)”, the feasibility of purifying total saponins with D101 macroporous resin instead of Amberlite-XAD-2 macroporous resin was investigated. The results showed that the standard curve showed good linearity in the range of 0 ~ 0.176 mg / m L with linear regression equation y = 3.308 4x-0.006 8 (R2 = 0.999 0). The average recovery of the improved method was 99.76%, RSD was 2.23% (n = 6), RSD of precision test was 2.97% (n = 6). The improved method of total saponins detection has the advantages of high sensitivity, good accuracy and good reproducibility. It can be used to determine the content of total saponin in Maca. The content of total saponin in Maca is 3.81 ± 0.11 mg / g (n = 6).
