目的通过建立热毒所致的兔动脉粥样硬化(Atherosclerosis,AS)模型,研究黄连解毒汤(Huanglian Jiedu Decoction,HJD)对兔动脉粥样硬化及内皮祖细胞(Endothelial progenitor cells,EPCs)的影响。方法在相同实验条件下,随机将30只新西兰雄兔分为5组,各组均静脉注射LPS(Lipopolysaccharides,LPS)联合高脂饲料喂养建立动脉粥样硬化模型,药物干预治疗前后,分别测定兔血脂的变化情况;抽取兔骨髓进行单个核细胞诱导培养、鉴定,加入含药血清后进行内皮祖细胞功能测定;观察腹主动脉及心脏组织病理形态学改变,了解AS形成情况及药物干预效果。结果建模4周末兔血脂水平达到高脂状态,中、高剂量黄连解毒汤及通心络干预组8周末血脂水平较4周末明显下降;黄连解毒汤中、高剂量组、通心络组含药血清对内皮祖细胞功能有明显促进作用;病理形态学观察,黄连解毒汤中、高剂量组、通心络组有显著减轻动脉粥样硬化形成的程度。结论黄连解毒汤可能通过降低血脂水平、抑制炎症反应及增强内皮祖细胞功能来延缓或防止热毒所致动脉粥样硬化的形成,为临床动脉粥样硬化的治疗提供参考。
Objective To investigate the effect of Huanglian Jiedu Decoction (HJD) on atherosclerosis and endothelial progenitor cells (EPCs) in rabbits by establishing thermo-toxic model of atherosclerosis in rabbits. . Methods Thirty New Zealand male rabbits were randomly divided into five groups under the same experimental conditions. The rats in each group were given LPS (Lipopolysaccharides, LPS) and high fat diet to establish the model of atherosclerosis. Before and after the drug intervention, rabbits The changes of blood lipids were observed. Rabbit bone marrow was harvested for induction and culture of mononuclear cells. The function of endothelial progenitor cells was assayed after adding serum containing drugs. The pathological changes of abdominal aorta and heart tissue were observed to understand the formation of AS and the effect of drug intervention. Results At the end of 4 weeks, the blood lipid level of rabbit reached high fat state. The levels of serum lipid of middle and high dose Huanglian Jiedu Decoction and Tongxinluo intervention group decreased significantly at the end of the 4th week compared with those at the end of 4th week. The middle and high dose Huanglian Jiedu Decoction group, Tongxinluo group Pharmacological serum could obviously promote the function of endothelial progenitor cells. Pathological observation showed that Huanglian Jiedu Decoction medium, high dose group and Tongxinluo group could significantly reduce the degree of atherosclerosis. Conclusion Huanglian Jiedu Decoction may delay or prevent the formation of atherosclerosis caused by thermo-toxic by lowering blood lipid level, inhibiting the inflammatory reaction and enhancing the function of endothelial progenitor cells, which may provide a reference for the clinical treatment of atherosclerosis.
