目的:探讨石斛对胃热证大鼠的作用及其机制。方法:用干姜水煎剂连续灌胃大鼠15 d,复制胃热证大鼠模型,再以石斛水煎剂(1.5,0.75 g.kg-1)灌胃治疗10 d。比较大鼠的饮食量、大小便、舌色舌苔变化,用红外热扫描成像(therm altexture m aps,TTM)系统检测大鼠全身不同部位的温度改变,用放射免疫分析法测定大鼠血液中与胃机能相关的生化指标血栓素B2(TXB2)、6-酮-前列腺素F1α(6-keto-PGF1α)、胃动素(MTL)、胃泌素(Gas)、生长抑素(SS)、白细胞介素-4(IL-4)、白细胞介素-8(IL-8),观察大鼠胃黏膜组织病理学变化情况。结果:造模后大鼠呈现舌红苔黄(P<0.05)、饮食量增加(10%以上)、大小便量减少(10%以上)、大便多数干结、小便颜色加深(P<0.01),大鼠的头、颈、左腋下、胸、上腹、中腹部体温显著升高(差值>0.5℃或差值>1.0℃);大鼠血液中显著降低的有6-keto-PGF1α(P<0.01),显著升高的有MTL,Gas,IL-8(P<0.01);大鼠胃黏膜受到损伤,呈现炎性变化(P<0.05,P<0.01)。石斛治疗后除食量,头、颈、胸部温度,血浆MTL浓度,胃黏膜腺体主细胞数外,上述胃热证的其他指标均得到改善(差值>0.5℃或差值>1.0℃,P<0.05,P<0.01)。结论:石斛通过使胃热证大鼠血液中的IL-8,Gas,TXB2降低,6-keto-PGF1α升高,改善胃热证大鼠的全身体征,发挥对大鼠胃黏膜的保护作用。
Objective: To investigate the effect and mechanism of Dendrobium on stomach heat syndrome rats. Methods: The rats were gavaged with Ginger Decoction for 15 days. The rat model of Genshen syndrome was replicated. The rats were treated with Dendrobium decoction (1.5,0.75 g.kg-1) for 10 days. The changes of diet, urine and tongue color in rats were compared. The changes of temperature in different parts of the body were detected by using the system of thermomechanical alterations (TTM). The radioimmunoassay Gastric-related Biochemical Indicators Thromboxane B2 (TXB2), 6-keto-PGF1α, motilin (MTL), gastrin, somatostatin (SS), leukocyte (IL-4) and interleukin-8 (IL-8) in gastric mucosa were observed. The pathological changes of gastric mucosa were observed. Results: After the model was made, the red tongue was yellow in the rats (P <0.05), the diet increased (more than 10%) and the amount of stool was reduced (more than 10% Rats in the head, neck, left armpit, chest, upper abdomen, mid-abdomen was significantly elevated body temperature (difference> 0.5 ℃ or difference> 1.0 ℃); significantly decreased blood in rats with 6-keto-PGF1α P <0.01). Significantly increased MTL, Gas, IL-8 (P <0.01). Gastric mucosa of rats were damaged and showed inflammatory changes (P <0.05, P <0.01). Dendrobium addition to food after treatment, head, neck, chest temperature, plasma MTL concentration, the number of main gastric mucosal glands, the other indicators of the stomach heat syndrome were improved (difference> 0.5 ℃ or difference> 1.0 ℃, P <0.05, P <0.01). CONCLUSION: Dendrobium can reduce the levels of IL-8, Gas, TXB2 and increase 6-keto-PGF1α in the blood of rats with stomach-heat syndrome, and improve the systemic signs of rats with stomach-heat syndrome and play a protective role on the gastric mucosa.
