将构建的2 种重组表达质粒pSV2uPA和pcDNA3upA分别直接注入小鼠骨骼肌,观察了ScuPAcDNA 在小鼠体内的表达情况- 体外凝血试验显示注射质粒DNA 后,第5 d 凝血时间有所延长,第15 d 凝血时间延长幅度最大,较对照组平均延长6 min 左右;第20 d 体内诱导产生DIC,体外凝血时间试验组较对照组延长一倍,抗凝效果明显,2 种表达质粒在体内的表达水平无明显差异-
The recombinant plasmids pSV2uPA and pcDNA3upA were directly injected into mouse skeletal muscle respectively to observe the expression of ScuPAcDNA in mice. In vitro coagulation test showed that after the plasmid DNA was injected, the 5th day clotting time After 15 days, the clotting time was prolonged by a maximum of 6 minutes compared with that of the control group. On the 20th day, DIC was induced in vivo. Compared with the control group, the clotting time in vitro was doubled and the anticoagulant effect was obvious.
