目的建立超高效液相色谱-串联质谱法测定黄芪中黄芪甲苷的含量。方法黄芪经甲醇-氨水(3:1,V/V)超声提取30 min,采用多反应监测(MRM)模式,以m/z 785.4/143.108和m/z 785.4/473.391为定性离子对,m/z 785.4/143.108为定量离子对,外标法测定黄芪甲苷。结果黄芪甲苷在60.0~12 000 ng/ml浓度范围内有良好线性关系(r=0.999 3),平均回收率为91.7%,最低检测限为68.2μg/kg。结论该方法具有简便、快速和灵敏度高的特点,可用于黄芪中黄芪甲苷的测定。
OBJECTIVE To establish a method for the determination of astragaloside Ⅳ in Radix Astragali by ultra performance liquid chromatography-tandem mass spectrometry. Methods Astragalus membranaceus was extracted by methanol - aqueous ammonia (3: 1, V / V) for 30 min and the m / z was determined by multiple reaction monitoring (MRM) with m / z 785.4 / 143.108 and m / z 785.4 / 473.391. z 785.4 / 143.108 for quantitative ion pairs, external standard method for the determination of astragaloside. Results Astragaloside had a good linearity (r = 0.999 3) within the range of 60.0-12 000 ng / ml with an average recovery of 91.7% and a minimum detectable limit of 68.2 μg / kg. Conclusion The method is simple, rapid and sensitive features, can be used for the determination of Astragaloside Astragaloside.
