目的 探讨牛磺酸对染锰大鼠脑损伤的保护作用.方法 选取Wistar大鼠45只,随机均分为染锰组、治疗组和对照组.对照组腹腔注射生理盐水30mg/kg;染锰组腹腔注射MnCl2·4H2O溶液15mg/kg,每天1次,每周5次,连续4周;治疗组在染锰组基础上,经皮下注射牛磺酸125.15mg/kg,隔天1次,连续10次.实施Morris水迷宫实验和跳台实验,连续测试4d.ELISA法测定大鼠丘脑谷氨酸酶的含量,黄嘌呤氧化酶法检测丘脑超氧化物歧化酶(SOD)含量,硫代巴比妥酸法检测丙二醛(MDA)含量.结果 染锰组Morris水迷宫实验错误次数和潜伏期以及跳台实验潜伏期均比对照组延长(P<0.05).治疗组上述3项指标均比染锰组缩短(P<0.05).染锰组谷氨酰胺酶、谷氨酸脱羧酶、谷氨酰胺合成酶和SOD均比对照组降低,MDA比对照组增高(P<0.01);治疗组上述5项指标变化均比染锰组少(P<0.01).结论 牛磺酸可通过调节谷氨酸代谢保护染锰大鼠的神经损伤,提高学习记忆能力.“,”Objective To investigate the protective effect of taurine on manganese-induced brain damage in rats.Methods Forty-five Wistar rats were randomly divided into three groups with 15 rats each.The rats in group C were injected abdominally normal saline 30 mg/kg as the controls.The rats in group A were injected abdominally MnCl2 · 4H2O solution 15 mg/kg daily for 5 days a week for 4 weeks.On the basis of MnCl2 · 4H2O injection as in group A,the rats in group B were injected subcutaneously taurine 125.15 mg/kg every other day for 10 times.Morris water maze test and platform test were performed daily for 4 days.The enzyme-linked immune method was used to detect the content of glutamic acid enzyme in the hypothalamus,xanthine oxidase method was used to detect the content of super oxide dismutase(SOD) and glucosinolates barbituric acid method was used to detect the content of malondialdehyde(MDA).Results Compared to group C,the number and incubation of Morris water maze error and incubation of step-down test in group A were prolonged (P<0.05),which were shorter in group B than those in group A(P<0.05).Compared to group C,transglutaminase,glutamic acid decarboxylase,glutamine synthase and SOD were lower,but MDA was higher in group A (P<0.01),the changes of which were less in group B than those in group A (P<0.01).Conclusion Taurine treatment may protect the manganese-induced brain damage and improve the ability of learning and memory in rats via regulating glutamate metabolism.
