探索成骨细胞的体外培养方法 ,建立人骨膜源性成骨细胞体外实验模型。方法 :以人髂骨骨膜为材料 ,通过植块培养法进行细胞培养 ,通过形态学观察 ;细胞的碱性磷酸酶 (ALP)染色 ;培养液中骨钙素 (BGP)的放免分析等进行细胞学鉴定。结果 :成骨细胞为成纤维细胞型 ,贴壁生长 ,形态多样化 ,HE染色细胞轮廓清晰 ,核内可见 2 -3个核仁 ;细胞的ALP染色为阳性 ;培养液中可检测出较高浓度的BGP。结论 :可以利用人骨膜为材料进行成骨细胞的体外培养 ,本培养方法 ,简便易行 ,可靠。
Explore osteoblast in vitro culture methods, the establishment of human periosteal derived osteoblasts in vitro experimental model. Methods: Human ilium periosteum was used as material, and cell culture was performed by explant culture method. Cell morphology was observed by alkaline phosphatase (ALP) staining and radioimmunoassay (RIA) of osteocalcin (BGP) Learn to identify. Results: Osteoblasts were fibroblast type with adherent growth and diverse morphology. HE-stained cells showed clear outline with 2-3 nuclei in nucleus. ALP staining of cells was positive. Concentration of BGP. Conclusion: Human periosteum can be used as the material to culture osteoblasts in vitro. The culture method is simple, easy and reliable.
