目的探讨天冬提取物作为流感疫苗佐剂的免疫增强作用,并应用二次回归正交旋转组合方法优化天冬佐剂与疫苗剂量。方法将不同剂量的天冬提取物与H3N2流感灭活疫苗共同肌肉免疫小鼠,以相应剂量灭活疫苗的单独免疫组、灭活疫苗与氢氧化铝共同免疫组、生理盐水免疫组作为对照组,间隔3周,免疫2次,取尾血收集血清,测定血凝抑制(HI)抗体效价。并且应用二次回归正交旋转组合方法,以天冬提取物及流感疫苗血凝素(HA)含量作为2个关键因素设计分组,将HI效价作为响应值,使用SAS9.1软件进行数据分析,对天冬提取物及疫苗剂量组合进行研究,并得出最佳响应值对应的二者剂量。结果天冬提取物能显著增强血清HI抗体效价,并且高于氢氧化铝佐剂。当流感疫苗HA含量为4.345μg、天冬提取物含量为0.1 mg时能够获得最高响应值(HI效价为5 092)。结论天冬提取物对流感灭活疫苗具有佐剂效果,并有望成为流感灭活疫苗的新型免疫佐剂。
Objective To investigate the immunopotentiation effect of Astellas sinensis extract as an adjuvant for influenza vaccine and to optimize the dose of Asparagus adjuvant and vaccine by quadratic regression orthogonal rotation combination method. Methods The mice were immunized intramuscularly with different dosages of asparagus extract and H3N2 influenza inactivated vaccine, the immunized group with corresponding doses of inactivated vaccine, the co-immunized vaccine with aluminum hydroxide and the saline-immunized group as control group , 3 weeks apart, immunized twice, the tail blood was collected to measure the titer of HI. And quadratic regression orthogonal rotation combination method, aspartate extract and influenza vaccine hemagglutinin (HA) content as two key factors in the design group, the HI titer as a response value, using SAS9.1 software for data analysis , The asparagus extract and vaccine dose combinations were studied and the best response values were obtained for both doses. Results asparagus extract can significantly enhance serum HI antibody titers, and higher than aluminum hydroxide adjuvant. The highest response (HI titer of 5 092) was obtained when the HA content of the influenza vaccine was 4.345 μg and that of asparagus extract was 0.1 mg. Conclusion Asparagus extract has an adjuvant effect on influenza inactivated vaccine and is expected to be a novel immunological adjuvant for influenza inactivated vaccine.
