目的 :探讨急性化脓性中耳炎时中耳粘膜发生的热休克反应。方法 :向豚鼠中耳注射肺炎克雷伯杆菌 ,制成中耳急性感染动物模型 ,分别于接种后第 1、3、5、7天处死动物取材。应用免疫转印技术动态分析中耳粘膜热休克蛋白 ( HSP)的表达。结果 :正常非应激状态下 ,豚鼠中耳粘膜仅有很弱的 HSP70的表达 ;中耳炎急性期 ,中耳粘膜 HSP70的表达明显升高 ,以第 3天为最强 ,第 3天开始表达 31k D分子 ,出现两条带 ;第 5天最强 ,至第 7天消失。 17k D处第 1~ 5天均有两条带 ,第 3天最高 ,第 7天减弱。结论 :观察发现 ,肺炎克雷伯杆菌感染豚鼠中耳后 ,中耳粘膜出现热休克反应 ,表达 HSP70及与之有相关表位的 HSP31k D和 HSP17k D分子
Objective: To investigate the heat shock response of middle ear mucosa in acute suppurative otitis media. Methods: The Klebsiella pneumoniae was injected into the middle ear of guinea pig to make the animal model of acute infection of the middle ear. The animals were sacrificed on the 1st, 3rd, 5th and 7th days after inoculation respectively. Dynamic analysis of middle ear mucosal heat shock protein (HSP) expression using immunotransfer technique. Results: Normal non-stressed guinea pig middle ear mucosa had only a weak expression of HSP70. In acute stage of otitis media, the expression of HSP70 in middle ear mucosa was significantly increased, which was strongest on the third day and 31k on the third day D molecules, there are two bands; the strongest on the fifth day, to disappear on the seventh day. 17k D at the first 5 days have two bands, the highest on the 3rd day, the seventh day weakened. CONCLUSION: The results showed that after Klebsiella pneumoniae infected the middle ear of guinea pig, the heat shock response appeared in the middle ear mucosa, HSP70k and HSP17kD molecules
