[目的]观察桦褐孔菌多糖对脂多糖(LPS)刺激小鼠巨噬细胞株Raw264.7细胞与小鼠腹腔巨噬细胞促炎性细胞因子表达的影响.[方法]利用桦褐孔菌多糖(40mg/L)处理被LPS(100μg/L)刺激的Raw264.7细胞株和小鼠腹腔巨噬细胞,采用半定量RT-PCR方法测定促炎性细胞因子肿瘤坏死因子(TNF)-α和白细胞介素(IL)-1βmRNA的表达;采用ELISA法测定TNF-α和IL-1β水平.[结果]与给予LPS刺激相比较,给予桦褐孔菌多糖和LPS同时刺激的Raw264.7细胞株和小鼠腹腔巨噬细胞促炎性细胞因子TNF-α和IL-1βmRNA表达水平均明显降低(P<0.05);给予桦褐孔菌多糖和LPS同时刺激的Raw264.7细胞株在48 h后与只给予LPS刺激相比较,细胞所分泌的促炎性细胞因子TNF-α和IL-1β水平均明显下降(P<0.05).[结论]桦褐孔菌多糖对LPS刺激Raw264.7细胞株和小鼠腹腔巨噬细胞促炎性细胞因子TNF-α和IL-1βmRNA表达水平及Raw264.7细胞株促炎性细胞因子TNF-α和IL-1β的分泌均有抑制作用.
[Objective] To observe the effects of Inonotrophus polysaccharides on lipopolysaccharide (LPS) -stimulated mouse macrophage cell line Raw264.7 and mouse peritoneal macrophages proinflammatory cytokines expression. [Methods] Raw264.7 cells stimulated with LPS (100μg / L) and mouse peritoneal macrophages were treated with polysaccharides (40mg / L), and the levels of proinflammatory cytokine tumor necrosis factor (TNF) -α And IL-1βmRNA were detected by enzyme linked immunosorbent assay (ELISA). [Results] Compared with LPS stimulation, Raw264.7 cells stimulated with polysaccharide from Lonicera macrophylla and LPS simultaneously The mRNA expression levels of pro-inflammatory cytokines TNF-α and IL-1β in peritoneal macrophages of strain and mouse were significantly decreased (P <0.05). Raw264.7 cells stimulated with polysaccharide of Lonicera macrophylla and LPS at 48 h The levels of TNF-α and IL-1β secreted by cells were significantly decreased after LPS stimulation (P <0.05). [Conclusion] The polysaccharide of Inonotus obliquus could inhibit the growth of Raw264.7 cells induced by LPS The levels of proinflammatory cytokines TNF-α and IL-1βmRNA in strain and mouse peritoneal macrophages and the expression of pro-inflammatory cytokines TNF- And secretion of IL-1β is inhibited.
