以7-苯乙酰胺基-3-氯甲基头孢烷酸对甲氧基苄酯(GCLE)为原料合成了7-苯乙酰氨基-3-(丙-1-烯基)-3-头孢烯-4-羧酸-4-甲氧基苄酯(GPRE),并对Wittig反应进行了研究。结果标明:氯化锂存在时,在v(二氯甲烷/m L)∶v(N,N-二甲基乙酰胺/m L)∶w(Wittig试剂/g)=3∶2∶1、反应温度-10℃的条件下,GPRE Z、E比值超过13.5,主峰(Z+E)超过97.0%。
7-Phenylacetamido-3- (prop-1-enyl) -3-cephem was synthesized from 7-phenylacetamido-3-chloromethylcephalosporanic acid p-methoxybenzyl ester (GCLE) -4-carboxylate (GPRE), and the Wittig reaction was studied. The results indicate that the molar ratio of v (methylene chloride / m L): v (N, N-dimethylacetamide / m L): w (Wittig reagent / g) When the reaction temperature is -10 ℃, the GPRE Z and E ratios exceed 13.5 and the main peak (Z + E) exceeds 97.0%.
