目的:探讨贯叶金丝桃中金丝桃素的最佳酶提取工艺。方法:以高效液相色谱法测定金丝桃素含量为指标,从纤维素酶、果胶酶、木聚糖酶、β-葡聚糖酶及复合酶SPE-007A中筛选出最优酶,采用单因素试验和正交试验优选提取工艺条件。结果:最优酶为纤维素酶,优选酶解工艺为酶用量2.0%,pH 4.8,温度50℃,酶解时间5 h,浸提倍数8倍。结论:该优选工艺高效、稳定。
Objective: To explore the best enzyme extraction technology of hypericin in Hypericum perforatum. Methods: The content of hypericin was determined by HPLC, and the best enzyme was screened from cellulase, pectinase, xylanase, β-glucanase and compound enzyme SPE-007A. Single factor test and orthogonal experiment were used to optimize the process conditions. Results: The best enzyme was cellulase. The optimal enzymatic hydrolysis process was enzyme 2.0%, pH 4.8, temperature 50 ℃, enzymolysis time 5 h and extraction ratio 8 times. Conclusion: The preferred process is efficient and stable.
