目的研究乌灵菌粉对大鼠戊四唑点燃癫痫形成过程的作用和慢性癫痫诱发记忆障碍的作用及机制。方法将SD大鼠随机分为4组:戊四唑点燃模型组(模型对照组),低剂量乌灵菌粉(0.3 g.kg-1,ig)组,高剂量乌灵菌粉(0.6 g.kg-1,ig)组和空白对照组。各组大鼠隔日腹腔注射亚惊厥剂量(35 mg.kg-1)的戊四唑(空白对照组注射生理盐水),共12次。乌灵菌粉在每次注射戊四唑前0.5 h灌胃。采用穿梭箱被动回避试验测定大鼠的记忆能力,化学荧光法测定脑内组胺含量。结果乌灵菌粉能延缓大鼠戊四唑点燃癫痫的形成过程,但不能阻断点燃的最终形成。戊四唑点燃癫痫使大鼠在穿梭箱中的被动回避时间缩短,乌灵菌粉明显逆转了这种被动回避时间的缩短。戊四唑点燃癫痫使大鼠脑内组胺含量明显下降,乌灵菌粉部分逆转了这种下降,在海马和下丘脑达到了统计学显著性意义。结论乌灵菌粉能延缓大鼠戊四唑点燃癫痫的形成过程,改善癫痫诱发的记忆障碍,其作用可能与增强脑内组胺能神经的功能有关。
Objective To study the effect of Wulin powder on the process of pentylenetetrazole-induced epilepsy in rats and the mechanism of memory impairment induced by chronic epilepsy. Methods Sprague-Dawley rats were randomly divided into four groups: pentylenetetrazole (model control group), low dose of Ugandan powder (0.3 g.kg-1, ig) kg-1, ig) group and blank control group. Rats in each group were injected intraperitoneally with pentapentazole at a dose of 35 mg · kg -1 (control group injected with saline) every other day for 12 times. Wuling powder 0.5 h before each injection of pentylenetetrazol gavage. The passive avoidance test of shuttle was used to measure the memory ability of rats, and the content of histamine in the brain was determined by chemiluminescence method. Results Wu Ling powder can delay pentylenetetrazole-induced epileptic seizures in rats, but can not interrupt the final formation of ignition. Pentylenetetrazole ignited epilepsy rats in the shuttle box passive avoidance time shortened, Wu Ling powder significantly reversed this passive avoidance time shortened. Pentylenetetrazole ignited epilepsy in rats brain tissue histamine decreased significantly, Wu Ling powder partially reversed this decline in the hippocampus and hypothalamus reached a statistically significant significance. Conclusion Wulin powder can delay pentylenetetrazole-induced epileptic seizures in rats and improve the memory impairment induced by epilepsy, which may be related to the enhancement of the function of histaminergic neurons in the brain.
