采用紫外分光光度仪测定茄尼醇的紫外透光率、体外清除自由基和酪氨酸酶活性抑制率,从三个方面探讨茄尼醇抗氧化和抑制酪氨酸酶的作用机制。观察了茄尼醇的最小紫外透光率及紫外透光率的日变化规律,参照Smirnoff反应方法建立反应体系模型来检测对羟基自由基(·OH)的清除力,采用邻苯三酚自氧化法检测对超氧阴离子自由基(O2-·)的清除力,还利用酶动力法研究了茄尼醇对酪氨酸酶活性的抑制作用。实验结果表明:茄尼醇能有效吸收紫外辐射,有效清除脂质自由基而阻断脂质过氧化,以及有效抑制酪氨酸酶。茄尼醇为天然提取物,可望用于抑制皮肤衰老及老年斑形成。
The ultraviolet transmittance of solanesol and the inhibition rate of free radical and tyrosinase in vitro were determined by ultraviolet spectrophotometer. The mechanism of anti-oxidation and tyrosinase inhibition of solanesol was discussed from three aspects. The diurnal variation law of minimum ultraviolet light transmittance and ultraviolet light transmittance of solanesol was observed. The reaction system model was established according to the Smirnoff reaction method to detect the scavenging ability of hydroxyl radical (· OH). The pyrogallol autoxidation Method was used to detect the scavenging ability of superoxide anion radical (O2- ·). The inhibition of solanesol on tyrosinase activity was also studied by enzyme kinetic method. The experimental results show that solanesol can effectively absorb ultraviolet radiation, effectively remove lipid free radicals and block lipid peroxidation, and effectively inhibit tyrosinase. Solanesol is a natural extract, is expected to be used to inhibit skin aging and senile plaque formation.
