目的:检测鸡卵清蛋白上游启动子转录因子Ⅱ(COUP-TFⅡ)在人胃癌组织中的表达,并探讨其临床意义及其影响胃癌发生、发展的可能分子机制。方法:收集66例手术切除的胃癌组织和相对应的癌旁非肿瘤组织标本,采用免疫组织化学法检测标本中COUP-TFⅡ和基质金属蛋白酶-2(MMP-2)蛋白的表达,分析COUP-TFⅡ与患者临床病理特征及预后之间的关系,以及COUP-TFⅡ与其潜在靶基因MMP-2的相关性。结果:COUP-TFⅡ蛋白在胃癌和癌旁组织中的阳性表达率分别为65.2%(43/66)和19.7%(13/66),两者比较差异有统计学意义(P<0.05)。COUP-TFⅡ蛋白的表达与胃癌的浸润深度、分化程度、淋巴结转移及TNM分期密切相关(P<0.05),而与患者的性别、年龄、肿瘤大小无关(P>0.05)。COUP-TFⅡ蛋白阳性表达的患者术后平均生存时间比阴性表达者短,分别为(26.8±2.6)个月和(36.4±3.0)个月(χ2=4.118,P=0.042)。MMP-2蛋白在胃癌和癌旁组织中的阳性表达率分别为78.8%(52/66)和34.8%(23/66),两者比较差异有统计学意义(P<0.05)。COUP-TFⅡ与MMP-2蛋白在胃癌组织中的表达呈正相关(r=0.310,P=0.011)。结论:COUP-TFⅡ蛋白与胃癌的发生、发展、侵袭、转移以及不良预后密切相关,推测可能是通过调控MMP-2的表达而发挥作用。
OBJECTIVE: To detect the expression of COUP-TFⅡ upstream of human egg ovalbumin in human gastric carcinoma and to explore its clinical significance and possible molecular mechanisms that affect the occurrence and development of gastric cancer. Methods: 66 cases of resected gastric cancer tissues and corresponding non-cancerous non-tumor tissue specimens were collected. The expressions of COUP-TFⅡ and MMP-2 protein were detected by immunohistochemistry. The expressions of COUP- The relationship between TFⅡ and the clinicopathological characteristics and prognosis of patients, and the relationship between COUP-TFⅡ and its potential target gene MMP-2. Results: The positive rates of COUP-TFⅡ protein in gastric cancer and paracancerous tissues were 65.2% (43/66) and 19.7% (13/66), respectively. The difference was statistically significant (P <0.05). The expression of COUP-TFⅡwas closely related to the depth of invasion, differentiation, lymph node metastasis and TNM stage (P <0.05), but not to the gender, age and tumor size (P> 0.05). The average survival time of COUP-TFⅡpositive patients was shorter than that of negative ones (26.8 ± 2.6) months and (36.4 ± 3.0) months respectively (χ2 = 4.118, P = 0.042). The positive rate of MMP-2 protein in gastric cancer and paracancerous tissues was 78.8% (52/66) and 34.8% (23/66), respectively. The difference between the two groups was statistically significant (P <0.05). COUP-TFII and MMP-2 protein expression in gastric cancer was positively correlated (r = 0.310, P = 0.011). CONCLUSION: The COUP-TFⅡ protein is closely related to the occurrence, development, invasion, metastasis and poor prognosis of gastric cancer, suggesting that COUP-TFⅡ may play a role by regulating the expression of MMP-2.
